  INHIBITORY EFFECT OF VARIOUS SOLVENT EXTRACTS OF FICUS CARICA FRUIT ON PROMYELOCYTIC LEUKEMIA CELL LINE (HL-60)

    ABSTRACT


    Background: Leukemia is one of the leading causes of death worldwide. The number of cases of leukemia is increasing over the half century, particularly among the males. There are various therapeutic options to treat this type of cancer but most of them are highly toxic. However, plants and plant products have been recognized as alternative sources for treatment and management of various ailments as they contained many bioactive compounds. Objectives: Thus, this study was conducted to investigate the inhibitory effects of the various solvent extracts (ethanol, ethyl acetate & hexane extracts) of Ficus carica fruit on human promyelocytic leukemia cells line (HL-60). Methods: Various solvent extracts of Ficus carica fruit were prepared through sequential maceration. The in-vitro inhibitory effects of different doses of the solvent extracts of Ficus carica fruit on promyelocytic leukemia cells was determined by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT) assay, for 72 hours. Results: All the solvent extracts of Ficus carica fruit inhibited the proliferation of promyelocytic leukemia cells. From the findings, 0.78125 µg/mL was the lowest concentration that inhibited cell proliferation. Ethyl acetate extract also strongly inhibited 50% of promyelocytic leukemia cells proliferation at concentration of 25 µg/mL. Conclusion: This study shows that solvent extracts obtained from Ficus carica fruit could inhibit the growth of promyelocytic leukemia cells. Furthermore, Ethyl acetate extract exhibited potent inhibitory effects via cytotoxic activities. The findings suggest promising results on the chemotherapeutic potential of Ficus carica fruit extracts. Further studies are recommended to isolate bioactive compounds of the solvent extracts of Ficus carica fruit in order to establish their inhibitory effects and mechanism of action.
